1尾仔鱼/0.05?0.5ml无水乙醇  一种提取单个鱼卵仔鱼微量总dna的简便快速方法  其特征在于:1)样品固定:采集鱼卵或仔鱼  滤去水后立即加入无水乙醇  2)样品中固定液的去除:取步骤1)所得已固定的单个鱼卵或仔鱼样品  而后于?20℃或室温保存备用  加入生理盐水洗涤  3)样品消化:取上述洗涤过的单个鱼卵或仔鱼  鱼卵或仔鱼与无水乙醇比例为:1ml鱼卵/2?5ml无水乙醇  1000?2000g离心20?60秒去除多余生理盐水  加入100?300μl?dna提取缓冲液和10?30μl细胞裂解缓冲液  4)提取总dna:将步骤3)消化至澄清的溶液中加入100?300μl?5?6mol/l?nacl溶液  再用滤纸吸干样品  使细胞破壁  震荡20?30秒。15000g离心30分钟  重复2?3次  而后将细胞破壁的单个鱼卵或仔鱼加入10?100μg蛋白酶k和10?100μg?rna酶  吸取上清液使蛋白质去除  震荡混匀  然后在上清液中加入等体积异丙醇  于50?70℃温育  轻轻混匀  每10?20分钟颠倒混匀一次  ?20℃静置1?2小时  消化至溶液澄清  15000g离心15分钟  待用  沉淀即得到单个鱼卵仔鱼微量总dna。
