诱导后皱纹盘鲍体液因子活性变化的初步研究以及抑菌因子的分离及其活性研究 | |

摘要 | 通过注射副溶血弧菌于皱纹盘鲍外套膜和足部以及进行超声波处理鲍,得到的结果显示上述三种方法均为有效诱导方法,且注射弧菌于外套膜部位更为有效。选择粘液和内脏团作为检测部位,两者的抗菌、溶菌、抑菌及凝集素活力都可检测出,且粘液部位各指标的平均活力高于内脏团。皱纹盘鲍的凝集素可凝集鸡红细胞,但不能凝集兔红细胞,表明其具有种的专一性。动力学研究表明注射弧菌后抗菌、溶菌、抑菌活力及凝集素活力明显提高。溶菌活力在86h内呈逐步上升状态,而抗菌、抑菌和凝集素活力在注射大约62h后达到峰值。凝集素活力在前38h未表现出明显变化,但在以后大幅升高,86h回落。注射副溶血弧菌于鲍外套膜62h后收集鲍血淋巴,经沸水浴处理后进行Sephadex G-100层析,择抑菌活力最高峰进行PAGE纯化。结果表明通过注射弧菌并经沸水浴处理后,通过Sephadex G-100可从鲍血淋巴分离出三个蛋白峰,最小的峰三蛋白呈现出最高的抑菌活力。峰三蛋白再经PAGE分离得出三条带,分子量最小的带占主要部分。因为峰三蛋白具有抗热的性质,且分子量较小,抑菌活力高,符合抗菌肽的性质,推测峰三蛋白为抗菌肽类物质。实验初步确立鲍鱼体内的一些免疫指标,为通过免疫方法增强皱纹盘鲍抗病力提供科学依据。 |
其他摘要 | By injecting Vibrio parahaemolyticrs in the mantles and feet or ultrasonic irradiating abalone, we obtain the results that all the three methods are effective immune methods and infecting mantles is the most effective method. Choose the phlegm and the pluck as the experiment samples, the results show that antibacterial, bacteriolytic, bacteriostatic and lectin activities and these three activities in phlegm are higher than in pluck. Abalone lectin can agglutinate erythrocytes of chicken but not rabbit. The kinetics indicates that injecting Vibrio parahaemolyticrs the antibacterial, bacteriolytic, bacteriostatic and lectin activities rise clearly,. The bacteriolytic activity rise successively during the 86 hours, but the bacteriostatic, antibacterial and lectin activities reach peak values at about 62 hours after injection. The lectin activity has no obviously change in the former two days, and rise a rather extent in the third day. We collect the haemolymph of abalone after injecting Vibrio parahaemolyticrs 62h, put it over seething water and through Sephadex G-100 chromatographic. Check their bacteriostatic activities, and choose the higher part do PAGE to purify it. The results show that after injecting Vibrio parahaemolyticrs and seething water bath, the haemolymph can be isolated into three parts by chromatography, the third part protein shows the highest bacteriostatic activity. And through PAGE, the third part protein is also isolated into three strips, and the major strip is the smallest one. Because by processed seething water bath, the third part protein has heatproof performance and high bacteriostatic activity, and is small, which corresponds with attacin's characters. It accounts that the third part protein is very possibly attacin. By this experiment we set some immune factors in abalone, and provide the proof for improving the defense ability of abalone. |
